For the Marriott Mega bonus promotion, does a single night stay counts as a “stay” or it needs to be more than one night?

Single night counts as a stay. All consecutive nights at a hotel count as one stay.

For promotions like these, when I want to stay several nights in the same city, I sometimes book a couple a nights at the hotel, stay at a different hotel for one night, then return to the original hotel. That counts as two stays.

The other aspect of Marriott is many locations have two different Marriott brand hotels next to each other like a Residence Inn and a Courtyard. Stay two consecutive nights with one night at each hotel and that counts as two stays.

When traveling with my wife I have booked one night in my name and next night in wife’s name. I check in and check out for our first night and she checks in and checks out for second night. We each earn one stay toward the promotion.

Repeat at a different hotel.

In 2003 we each earned 50,000 bonus Starwood points doing that for 10 consecutive nights at 5 hotels in Melbourne, Sydney and Noosa, Australia.
